An immunological method for quantization of Hevea Natural Rubber (HNRL) proteins using rabbit anti-HNRL serum. Rabbits immunized with HNRL proteins react to the majority of the proteins present, and their sera have the capability to detect most if not all the proteins in HNRL.
Scope:
ASTM D6499 quantitatively measures the level of antigenic latex proteins in solution using an inhibition format which detects antigenic proteins. It determines the amount of antigenic protein in Hevea Natural Rubber and its products using rabbit antisera specific for HNRL proteins.

Antigenic Protein determination in Hevea Natural Rubber by ASTM D6499 is done using Mooney Viscometer which involves calculation of three major components: Viscosity, Stress Relaxation, and Pre-Vulcanization Characteristics. ASTM D6499 deploys the shearing torque technique by resisting rotation of a cylindrical metal disk (or rotor) embedded in rubber within a cylindrical cavity. Testing phase includes material preparation, distribution, liaison with the participating laboratories and handling of test result data received from these laboratories.Evaluating test sensitivity for rubber covers the evaluation of chemical constituents, physical properties of compounding materials and cured rubbers. It describes how test sensitivity, generically defined as the signal-to-noise ratio, is evaluated in the rubber manufacturing industry.
