Spectrophotometry Testing Procedure

Spectrophotometry deals with the quantitative measurement of the reflection or transmission of light exhibited by a material as a function of wavelength. The measurements are done by an instrument known as the spectrophotometer, which measures the intensity of a light beam at different wavelengths. In other words, it measures the amount of light absorbed by a material. 

A spectrophotometer employs a spectrometer that produces light of different wavelengths and a photometer that measures light intensity. The specimen is placed in between the spectrometer and the photometer. As light passes through the specimen, certain wavelengths are absorbed. The amount of light that passes through the sample is measured by the photometer which sends a corresponding voltage reading to the display. 

Spectrophotometers are of two main types depending on the range of wavelength detected – IR and UV-visible spectrophotometers – though modern meters can also work with the electromagnetic spectrum. They are available in two variants as well – bench (stationary) and portable – both having their own unique applications. The main parts of a spectrophotometer include – a light source, a monochromator, a wavelength selector, a collimator, a photoelectric detector, a cuvette, and a digital display.
