ASTM E578-07 Linearity of Fluorescence Measuring Systems
ASTM E578-07 test method determines the properties of fluorescence measuring systems using low-energy excitation sources to detect the limits of the linearity of response with fluorescence intensity. The method is applicable for fluorescence measuring systems using a single detector. The SI units are considered to be the standard of measurement.
Scope:
ASTM E578-07 test method covers the performance properties of the fluorescence measuring systems. The limits of the linearity response are determined with respect to the range of concentration of a fluorescence substance. The stiffness of the material, recovery of the deformed portion, strain calculation, functioning, product assessment, transformation temperatures, quality control, specification acceptance, and research are dealt with in this method. This creates ease for the customer and supplier to reach an agreement.
Procedure:
The test apparatus uses a standard test solution of quinine sulfate dehydrate in sulfuric acid to test the linearity of fluorescence measuring systems. The instruments like cuvettes, slits, or filters are used to detect the fluorescence of the test solution in a measuring system. Multiple readings are taken, and a graph is plotted. The upper and lower limits of linearity are determined and expressed in micrograms per milliliter of the standard solution.
Specimen Size:
Specimen size should be as per the requirements. This test method can be applied to 10-mm path length cuvette formats and instruments covering a wavelength range within 190 to 900 nm.
Data:
The difference between the fluorescence intensity and the reading of the blank solution is represented by ‘S.’ ‘F’ represents the observed fluorescence intensity.
