Scope

This ASTM E1428 test method can be used to evaluate antimicrobials for staining by Streptoverticillium reticulum, and it helps to predict how well-treated items will perform in field conditions.

Procedure

To make a solidified agar layer for the ASTM E1428 test, enough yeast malt extract agar is poured into suitable sterile dishes. The agar surface is moistened by streaking the agar with a cotton swab dipped in water that is 3 mL sterile saline. Streptoverticillium cells are harvested. By streaking, the cell-laden wipe is applied to the agar surface. To place the sterile test specimens, sterile forceps are used.

The relationship between the test specimen and the infected agar is established. If the specimens are prone to curling, they are placed on top of the surface to keep the specimens in excellent contact. The test plates are covered with the infected bacteria. The specimens are incubated for 14 days.

Specimen size

The specimens should be 0.75 in. diameter.

Data

For the ASTM E1428 test, the degree of staining on the samples is observed. If no organisms have grown on the agar adjacent to the specimen, this is a “zone of inhibition.” This suggests that the antibiotic is effective.is possible that the specimen is leaching.
